Figure 1 — Schematic of a confocal laser, scanning microscope. Note that the rays representing fluorescence originating above and below the focal plane are blocked by the confocal aperture. By changing the focal plane, it is possible to section the sample optically. Two photon excitation offers intrinsic optical sectioning, thus eliminating the need for a confocal aperture.
